I purchased a Skymedi 2Gb USB 2.0 Flash drive. When I insert it into my PC (Core Duo 2, 2GB Ram, Windows XP Media edition) I get the message 'This disk is not formatted, do you wish to format now?' I click yes, but then get an error message, 'There is no disk in Drive E, please insert a disk'.

I have tried e-mailing skymedi to request a driver or whatever is necessary, but get no reply.

Any ideas how I can format the disk to work on my PC?

If this is a new drive I would return it to where you bought it. As a new drive it should not be like this. Some people have been able to get drives like this working again, but it is not always possible and they can revert to this state again in time. Basically, I think this is a faulty drive.
